纯化方法的不合理,主要是因为重组精氨酸脱亚胺酶一般是以无蛋白活性的包涵体形式表达,蛋白复性这一步就比较关键,报道过的纯化方法一般是先稀释复性然后再纯化,而稀释复性后的蛋白浓度很低,在浓缩蛋白时以及后面的纯化过程中由于精氨酸脱亚胺酶的不稳定性,很容易引起酶蛋白的活性损失。
